Triptorelinum is an antineoplastic hormone which is structurally analogous to natural gonadotropin releasing hormone (GNRH). By pharmacological activity it belongs to antitumor drugs, or suppressors of gonadotropins and cell division. Initially it makes pituitary gland to produce LH and FSH hormones. It causes temporary rise of estrogens and testosterone in the organism. But afterwards, the production of pituitary hormones (LH and FSH) falls, which leads to reduction of sex hormones.
Triptorelinum is stronger than endogenous hormone. It reaches the peak effect in three-four weeks when the rise of pituitary gonadotropins production comes to full stop. In this period the amount of estrogen produced by ovaries reaches the level specific for menopause, and the amount of testosterone – to the level of postcastration. When you discontinue using the substance, the level of endogenous hormones returns to norm.
Indications and contraindications
Triptorelinum is used to treat different hormone dependent disorders, such as prostate cancer, uterine myoma, hypogonadotropic amenorrhea, early puberty, endometriosis etc. Contraindications include: individual intolerance, prostate cancer which is not dependent on hormones, osteoporosis in women. As there are no adequate researches about the influence of the drug during pregnancy and breastfeeding, avoid using the substance during this period.
Bioavailability
It has very low bioavailability when taken orally. If you take it in depot form it may have prolonged action as the substance gradually dissolves from microcapsules and may keep the needed concentrations (200 – 500 ng/ml) for as long as thirty days after one injection. In case of intramuscular administration the bioavailability equals to 38% and in case of subcutaneous – 69%. The rate of degradation of Triptorelinum is 3 times slower than that of natural hormone.
